작용기전
[Polydeoxyribonucleotide(Defibrotide)]
Defibrotide는 섬유성 응괴(fibrin clots)를 가수분해하는 플라스민(plasmin) 효소 활성을 증가시킴. von Willebrand factor와 plasminogen activator inhibitor-1 발현을 감소시킬 뿐만 아니라 조직 plasminogne activator와 thrombomodulin 발현을 증가시켜 내피세포 활성화를 감소시키고 내피세포 매개 섬유소 용해를 증가시킴.

[Polydeoxyribonucleotide(Defibrotide)]
- 분포: Vd: 8.1-9.1L
- 단백결합률: 혈장 단백질에 93%가 결합됨.
- 대사: Polynucleotides는 nucleases, nucleotidases, nucleosidases, deaminases 및 phosphorylases에 의해 oligonucleotides, nucleotides, nucleosides로 대사된 후 유리 2’-deoxyribose sugar, purine 및 pyrimidine bases로 대사됨.
- 반감기: 2시간 미만
- 배설: 소변(총량의 5-15%)
